According to the initial reports, five coaches of the train have been derailed injuring about 42 passengers.
Police and railway officials said that the mishap occurred around 2 AM after five coaches of Chennai-Mangalore Express derailed at Puvanur in Vriddhachalam.
The injured have been rushed to nearby hospitals, official said, adding that the train services on the busy route were disrupted for sometime.
However, trains were later operated on alternative route but they were running late, official added.
"One person seriously injured, 3 persons received simple injuries and 35 injured were given medical aid on spot", said Anil Saxena, Chief PRO, Railways.
He further said, "Five rear coaches were derailed, so we will disconnect the affected coaches and take the train forward."
Officials have also said the Railway minister, Suresh Prabhu, is monitoring the situation.
This comes weeks after the twin rail derailment in Madhya Pradesh and highlights the creaking infrastructure and much needed investment required into the Indian Railways.
